From downtown Buffalo, head to the NFTA Metro Bus Terminal located at 181 Ellicott St. Take the #40 bus towards Fort Erie. The bus will take you across the Peace Bridge into Canada. Once you reach Fort Erie, disembark at the stop nearest to 350 Lakeshore Rd. Walk south on Central Ave and turn left onto Lakeshore Rd. Continue walking until you reach Old Fort Erie at 350 Lakeshore Rd.

If you have a bicycle, start from Canalside in downtown Buffalo. Head west along the waterfront to the entrance of the Niagara River Greenway Trail. Follow the trail north towards the Peace Bridge. After crossing the bridge, continue straight on Niagara Blvd, then turn right onto Central Ave. Finally, turn left onto Lakeshore Rd, and you'll find Old Fort Erie at 350 Lakeshore Rd.
