Turn left onto Calle Chihuampata, and you will soon see the Templo del San Blas ahead on your right. The walk should take about 10-15 minutes.

Buses heading to 'San Blas' are common, and you can catch them from various points in Cusco. You'll pay around 1-2 soles for the fare.
