From the main plaza in Manatí, head east on Avenida Manuel Corchado y Juarbe. Continue straight until you reach the intersection with Calle José de Diego. Turn left onto Calle José de Diego and walk until you reach the bus stop where local 'guaguas' (public vans) stop. Take a guagua heading to Jayuya. Inform the driver that you're going to Museo El Cemí. Once you arrive near Coabey, get off at the nearest stop and then walk approximately 10 minutes northeast on Route 143, following the signs to the museum.

From Manatí, locate the local bus terminal. Board a bus heading towards Jayuya. During your trip, kindly ask the bus driver to notify you when you reach the area closest to Museo El Cemí. Once you arrive, walk towards Route 143, and you will find the museum about a 10-minute walk to the northeast from the drop-off point.
