From the center of Ponce, head to the nearest bus terminal, which is called 'Terminal de Carros Públicos de Ponce' located at Calle Mendez Vigo. Once there, look for a bus heading towards Yauco. Buses to Yauco typically depart every 30 minutes. Inform the driver that you want to go to 'Iglesia Cristiana Ciudad del Gran Rey'. The bus will take approximately 30 minutes to reach Yauco. After arriving in Yauco, walk about 500 meters down Calle Manuel A Negrón, and you will find the church at number 48 on your right.
