From San Juan, take Route 52 south towards Ponce. Merge onto Route 2 west towards Mayagüez. Continue on Route 2 until you reach Route 100. Take Route 100 south towards Cabo Rojo. Follow signs for Cabo Rojo and then Route 301. Turn onto Route 301 and follow it until you reach the entrance of Refugio Nacional de Vida Silvestre de Cabo Rojo at Pole Ojea, Cabo Rojo 00622. The total drive is approximately 2 hours.

Take a public bus from San Juan to Ponce (bus number 21 or similar). Once in Ponce, transfer to a bus that heads towards Cabo Rojo (check for available services as they may vary).
