Take any bus that heads towards downtown Houston. Get off at the 'Main St @ Capitol St' stop. From there, transfer to the METRO light rail at the Main Street Square station. Board the 'Green Line' train towards NRG Park. Get off at the 'Museum District' station. After exiting the station, walk west on Burke Ave for a few blocks until you reach Waugh Dr. The bat colony is located near the bridge over Buffalo Bayou.
