From the main bus terminal in Maldonado, take the bus heading towards Punta del Este. Make sure to check the bus schedule for the latest times. After approximately 25 minutes, ask the driver to drop you off at the Punta Colorada stop. Once you get off, walk towards the beach; it should be a short 5-minute walk straight ahead.
Taxi
You can easily find a taxi at various locations in Maldonado, including the main square or your hotel. Inform the driver that you want to go to Punta Colorada Beach (4Q23+2M4, Punta Colorada). The ride will take around 20 minutes, depending on traffic.
