If you are driving from Zarafshan Range, head south on the M-37 highway towards Bukhara. Continue for approximately 150 kilometers until you reach the outskirts of Bukhara city. At the first major intersection, turn left onto Afrosiab Street (QCH5+4Q4). Follow Afrosiab Street for about 5 kilometers, and you will see the Ark of Bukhara fortress on your right. There is parking available near the entrance.
