From Sedona, head to the nearest bus stop on Highway 89A. You can use the Verde Lynx bus service. Take the Verde Lynx Route 1 towards Cottonwood. After approximately 30 minutes, you will arrive at the Flagstaff Transfer Station. From there, switch to the Mountain Line Bus Route 10 heading towards the Lowell Observatory. Get off at the Mars Hill Road stop, and the observatory will be a short walk uphill from there.

Bicycle
If you're feeling adventurous, you can rent a bicycle in Sedona. Head towards Highway 89A and follow the bike route that leads northeast out of Sedona towards Flagstaff. The ride will take about an hour and a half, depending on your pace. Once you reach Flagstaff, navigate to West Mars Hill Road. The observatory is at the end of this road, with clear signage to guide you.
