From Downtown Tucson, head to the nearest Sun Tran bus stop, which can be found at the corner of Congress St and 6th Ave. Take bus route 1 (the 'A' line) heading towards the Tucson Mall. You will need to transfer to route 99 at the Tohono Tadai Transit Center. Board route 99, which will take you directly to Old Tucson. The fare for a single ride is approximately $1.75, and be sure to check the latest schedule for the bus timings.
Bicycle
