Take tram number 3, 4, or 10 and get off at the 'Jiráskovo náměstí' stop. The church is a short 5-minute walk from the tram stop. Head northeast on Jiráskovo náměstí, then turn left onto V Jirchářích. The church will be on your right. A single ticket on public transport costs around 30 CZK and is valid for 90 minutes.
