If you are driving from any location within Big Cypress National Preserve, head southeast on US-41 E (Tamiami Trail). Continue for approximately 20 miles until you reach the intersection with FL-997 S. Turn left onto FL-997 S and continue for about 8 miles. You will need to turn right onto SW 344th St and follow it for 3 miles. The Southern Glades Wildlife and Environmental Area will be on your left. Look for the entrance sign.

To reach Southern Glades Wildlife and Environmental Area using public transportation, you can take a bus from the nearest transit station located near the Big Cypress National Preserve. First, take the bus route that heads towards Homestead, then transfer to a bus heading south on FL-997. Disembark at the nearest stop to SW 344th St. From there, you will need to walk approximately 1.5 miles east along SW 344th St to reach the entrance of Southern Glades Wildlife and Environmental Area. It is advisable to check the local transit schedules as they may vary.
