Once you reach Sarasota, take a right onto Fruitville Rd (FL-780) and follow it until you reach US-41. Turn left onto US-41 and then make a right onto Ken Thompson Parkway. Follow Ken Thompson Parkway for about 1 mile, and you will find Mote Marine Laboratory & Aquarium on your right at 1600 Ken Thompson Pkwy, Sarasota, FL 34236. Note that there may be parking fees of around $5-$10 depending on the time of year.
