Once you reach Reykjavík, take the exit for Hringbraut (Road 49). Continue on Hringbraut for about 3 km, and then turn left onto Efstaleiti. Perlan will be on your right after about 1 km. There is parking available at the site, but be aware that parking fees may apply.
