If you are traveling by car, start your journey from Johor Bahru. Head southwest on Jalan Wong Ah Fook towards Jalan Ibrahim. Take the ramp onto the North-South Expressway (E2). Continue on E2 for approximately 40 kilometers, then take exit 256 towards Pontian. Follow the signs to Kukup, which will lead you onto Jalan Kukup. After about 18 kilometers, you will arrive at Kukup Town. From Kukup Town, take a boat to Kukup Island Johor National Park. The boat ride takes about 20 minutes and typically costs around RM 15 per person. Ensure to check the boat schedules at the docks.

For those using public transportation, take a bus from Johor Bahru to Pontian. Buses such as the 'Causeway Link' or 'Transnasional' operate this route. The journey will take about 1 to 1.5 hours, and the fare is approximately RM 10 to RM 15. Once you arrive in Pontian, you can take a local taxi or a Grab car to Kukup Town, which is about 20 kilometers away. The taxi fare should be around RM 25. From Kukup Town, you can take a boat to Kukup Island Johor National Park, costing approximately RM 15 per person for a 20-minute ride.

If you prefer a more direct route, you can use a taxi or Grab car service from anywhere in Johor to Kukup Town. Make sure to confirm with the driver that they are familiar with the location of the boat terminal in Kukup Town.
