Walking
From the Brno Main Train Station (Hlavní nádraží), exit the station and turn left onto Masarykova street. Walk straight for about 500 meters until you reach the intersection with Česká street. Turn right onto Česká street. Continue walking for another 300 meters until you reach the square known as Náměstí Svobody. From the square, take the street called Minoritská, which is located to the southeast of the square. The Church of St. John will be on your left after approximately 200 meters, at Minoritská 449.

If you prefer to take public transport, from the Brno Main Train Station (Hlavní nádraží), head to the tram stop right outside the station. Take tram number 4 or 9 in the direction of the city center (centro). Get off at the 'Náměstí Svobody' stop. From there, walk southeast towards Minoritská street. The Church of St. John is located about 200 meters from the square at Minoritská 449.
