Car
From Uplistsikhe, take the main road (E117) towards Tbilisi. Drive for about 15 km until you reach the junction for Mtskheta. Take the exit towards Mtskheta and continue straight for approximately 8 km. You will see signs for Birtvisi Fortress. Follow the signs, which will lead you to a dirt road that requires a 4x4 vehicle. The fortress is located a short distance up this road. Be cautious as the road can be rough.

Public Transportation
From Uplistsikhe, you need to take a marshrutka (minibus) to Mtskheta. Marshrutkas depart frequently from the main road in Uplistsikhe. The fare is usually around 1-2 GEL. Once in Mtskheta, you can either take a taxi (approximately 20 GEL) directly to Birtvisi Fortress or walk to the main road and look for a local bus heading towards Tbilisi that stops near the fortress. Note that public transportation may not operate frequently, so check the schedules.
