Walking
If you are in the center of Assisi, start at the Basilica di San Francesco. Head east on Via San Francesco until you reach Piazza del Comune. From there, take Via Portica which will lead you to the main road. Continue straight until you see signs for Spoleto. Once you reach Spoleto, follow the signs to the city center where Piazza del Duomo is located. It should take approximately 25-30 minutes to walk.
