From the main square of San Gimignano, Piazza della Cisterna, head east towards Via San Giovanni. Continue straight until you reach the junction with Via Vecchia. Turn left onto Via Vecchia and walk for about 300 meters. You will come to a small bus stop on your right. This is where you will catch the bus to Pratolino. Make sure to check the local bus schedules for the times since buses may not run frequently. Once you board the bus, enjoy the scenic ride through the Tuscan countryside.
