If you are staying in the Centro area of San Miguel de Cozumel, head towards the main square (Plaza Central). From there, walk south on Avenida 5ta Avenida until you reach the waterfront. Keep walking until you reach the southern end of the island, where you will see the signs for the Zona Hotelera Sur. The Cozumel Trolley Tour & Co. is located at Carr. Costera Sur Km. 15, near notable landmarks such as the Parque Nacional Arrecifes de Cozumel.
