From Plaza de Armas in Cusco, head to the nearby bus terminal, known as 'Terminal Terrestre de Cusco', located on Avenida Huayruropata. Look for a minibus or colectivo (shared taxi) heading to Mollepata, which is the town nearest to the Humantay Lake Trek. The bus fare is usually around 10-15 soles, and the journey takes approximately 2-3 hours. Inform the driver that you need to get off at Mollepata.

Once you arrive in Mollepata, you will need to walk or take a local taxi to the trailhead for Humantay Lake. The trailhead is about 7 kilometers from Mollepata. You can ask locals for directions to the 'Humantay Lake Trek' starting point, which is well-marked. If you choose to walk, it will take about 1.5 to 2 hours to reach the trailhead.

Alternatively, upon arrival in Mollepata, you can hire a local taxi to take you directly to the trailhead. This is a more convenient option if you want to save time and energy. The taxi ride should cost around 30-50 soles and takes about 15-20 minutes.
