From any location in Hluhluwe-Imfolozi Park, head towards the N2 highway. Depending on where you are in the park, follow the signs directing you to the N2 South. Once on the N2, continue driving for approximately 40 kilometers until you reach the turn-off for the R618 towards St. Lucia. Follow the R618 which will lead you into the town of St. Lucia. Once in St. Lucia, follow the signs to Lake St. Lucia. The total journey will take around 1 hour, depending on traffic and road conditions.
