Bicycle Rental
If you're starting from downtown Key West, head to a nearby bike rental shop. From there, head east on Caroline Street until you reach the intersection with Whitehead Street. Turn left onto Whitehead Street and continue until you reach the end. You'll need to follow the road around the corner onto South Street, which will take you to US-1. From there, head south on US-1 for about 30 miles. The National Key Deer Refuge Nature Center will be on your right at 30587 Overseas Hwy.

You can take the Lower Keys Shuttle, which operates from Key West to Big Pine Key. Start at a nearby bus stop in Key West, and make sure to check the shuttle schedule for the next departure. The bus will take you directly to Big Pine Key. Once you arrive at Big Pine Key, the National Key Deer Refuge Nature Center is located at 30587 Overseas Hwy. It's a short walk from the bus stop.

Walking
Walking to the National Key Deer Refuge Nature Center from downtown Key West is quite a trek, and is recommended only for the adventurous. Start by heading north on Duval Street until you reach Truman Avenue. Turn left on Truman Avenue and continue until you reach US-1. Follow US-1 south for about 30 miles. Although this route is long, you can enjoy the scenic views along the way. The Nature Center will be on your right at 30587 Overseas Hwy.
