Walking
If you are starting from the Basilica of Santa Chiara, head south on Via Santa Chiara towards Piazza del Comune. Continue straight to Piazza del Comune and take a moment to admire the beautiful buildings around you. From here, take Via San Francesco which will lead you towards the main road. Once you reach the intersection with Via Giacomo Matteotti, turn left. Follow this road until you reach the bottom of the hill. When you reach the junction with Via Ugolino, turn right. Continue walking straight, and you will eventually reach Spello. The Roman Amphitheater is located at Via Centrale Umbra in Spello. It's approximately a 30-minute walk from Assisi.

From Assisi, head to the bus station located near the Basilica of San Francesco. Take a local bus towards Spello; the journey should take about 20 minutes. Once you arrive in Spello, disembark at the main bus stop. From there, walk along Via Centrale Umbra, and you will see the Roman Amphitheater within a 10-minute walk.

If you are near the Assisi train station, you can take a train to Spello. Check the schedule for trains heading towards Foligno and ensure that you get off at Spello station. The train ride takes about 5 minutes. After arriving at Spello, walk towards Via Centrale Umbra, and the Roman Amphitheater is about a 15-minute walk from the train station.
