From the Appalachian Mountains, head towards Interstate 81 (I-81) or Interstate 76 (I-76) depending on your starting point. If you're near Harrisburg, take I-76 East towards Philadelphia. If you're coming from the western part of the mountains, I-81 to I-76 East will be your route. Continue on I-76 until you reach the exit for I-95 North. Merge onto I-95 North. Take exit 22 for Callowhill St toward US-30 E. Continue onto Callowhill St, and then turn left onto 2nd St. Follow 2nd St until you reach Elfreth's Alley, and turn right onto it. The museum will be at 126 Elfreth's Alley, Philadelphia, PA 19106. Parking may be limited in the area, so be prepared to use a parking garage or street parking, which may incur fees.

If you prefer public transportation, you can take a Greyhound or Megabus from various locations in the Appalachian region to Philadelphia. Once you arrive at the bus station in Philadelphia, take the SEPTA (Southeastern Pennsylvania Transportation Authority) subway. Board the Market-Frankford Line at 69th St Terminal heading towards Frankford Terminal. Get off at the 15th St station and transfer to the Broad Street Line towards NRG Station. Get off at City Hall and walk east to 126 Elfreth's Alley, which takes about 10-15 minutes. Be sure to check the schedules for both bus and subway for any potential delays or service changes.
