If you are in the central plaza of Puno (Plaza de Armas), head towards the east side of the plaza where you will find the large stone buildings. Walk straight along Av. 28 de Julio for approximately 5 blocks until you reach the intersection with Jr. Ayacucho. Turn left onto Jr. Ayacucho, and continue walking for about 2 blocks. The Puno Cathedral will be on your right at Jr. Ayacucho 416.

From the main bus terminal in Puno, take a local mototaxi or shared taxi towards the city center (Plaza de Armas). Ask the driver to drop you off at Plaza de Armas. Once you arrive at the plaza, head east along Av. 28 de Julio for about 5 blocks. Turn left onto Jr. Ayacucho and walk for 2 blocks. The Puno Cathedral will be on your right at Jr. Ayacucho 416.
