If you're driving from Valdivia, take Route 5 towards Panguipulli. After about 60 km, take the exit towards Neltume on Route CH-203. Continue on CH-203 for approximately 25 km until you reach the entrance of Huilo Huilo Reserva Biológica at km 55. Look for signs along the way to ensure you're on the right track.
