If you are in the heart of Altstadt, head towards the famous Residenz Palace. From the Residenz, walk east along the Residenzstraße for about 10 minutes. After you pass the intersection with Juliuspromenade, continue straight until you reach Domstraße. Turn right onto Domstraße, and the Wurzburg Cathedral will be on your left at Domstraße 40.
