From the central Piazza della Cisterna in San Gimignano, head east on Via San Giovanni. Continue straight until you reach the intersection with Via Vecchia. Turn left onto Via Vecchia and follow it until you reach the end of the street. You will find a bus stop near the end of Via Vecchia, where you can catch a bus heading to Florence. Once you are on the bus, enjoy the scenic ride to Florence. It will take about an hour. When you arrive in Florence, get off at the main bus station, Firenze Santa Maria Novella. From there, you can either walk or take public transport to reach Chiesa di San Niccolò Oltrarno.

After getting off at Firenze Santa Maria Novella, exit the station and head southeast on Piazza della Stazione. Continue onto Via Panzani, which will become Via della Storia. Follow it until you reach Piazza della Repubblica, then turn left onto Via del Corso. Continue straight and cross the bridge at Ponte alla Carraia to reach the Oltrarno district. Once across the bridge, you will want to turn right onto Via di San Niccolò. Continue straight on this road, and you will find Chiesa di San Niccolò Oltrarno at Via di S. Niccolò, 75n.
