If you are near Shinjuku Station, take the Tokyo Metro Marunouchi Line (red line) and head towards Ogikubo. Get off at Nishi-Shinjuku Station, which is just one stop away. Once you arrive at Nishi-Shinjuku Station, take the exit A1. After exiting, turn left and walk straight for about 5 minutes. You will see the Tokyo Metropolitan Government Building on your left.

From Shinjuku Station's West Exit, you can take the Toei Bus (No. 01 or No. 02) towards 'Shinjuku Gyoenmae'. Get off at the 'Tokyo Metropolitan Government Building' stop. The building will be directly in front of you as you exit the bus. The bus ride takes about 10 minutes, depending on traffic.
