Car
From Punakha Town, head north on the Wangdue-Punakha Highway (NH 7). Continue for about 25 kilometers. As you approach Semtokha, keep an eye out for the signs directing you to Simtokha Dzong. Turn left onto Je Bamo Lam SE and you will find the Dzong about 1 kilometer down this road on your right.
Public Transportation
Take a local bus from Punakha Town heading towards Thimphu. Inform the driver that you want to get off at Semtokha. The bus will take you along the Wangdue-Punakha Highway (NH 7) for approximately 25 kilometers. Once in Semtokha, you can either walk towards Je Bamo Lam SE, which is about a 15-minute walk, or you can hire a local taxi to Simtokha Dzong, which is just 1 kilometer away.
Taxi
If you prefer to take a taxi, you can easily hire one from Punakha Town. Instruct the driver to take you to Simtokha Dzong via the Wangdue-Punakha Highway (NH 7). The ride will take approximately 30 minutes depending on traffic, and the driver will drop you off directly at the Dzong entrance.
