If you are starting from the main pier in Puerto Baquerizo Moreno, head northwest along Av. Charles Darwin. Continue straight for about 500 meters until you reach the intersection with Av. Isabella. Turn left onto Av. Isabella and continue walking for another 100 meters. Touring Galapagos will be on your right side, just before you reach the intersection with Calle La Lobería.
