From downtown Tucson, walk to the Ronstadt Transit Center located at 215 E Congress St. Board the Sun Tran Bus Route 19 towards 'Tucson Marketplace'. Ride for about 30 minutes until you reach the stop at 'W Congress St & S 6th Ave'. Transfer to Bus Route 34 towards 'Kinney Rd & E 36th St'. Ride for another 20 minutes. Get off at the 'Kinney Rd & Desert Museum' stop. From there, it's a short walk to the entrance of the Arizona-Sonora Desert Museum located at 2021 N Kinney Rd.

If you're comfortable biking, rent a bike from one of the many rental shops in downtown Tucson. Head west on W Congress St, then turn right onto S 6th Ave. Continue on S 6th Ave as it becomes W 22nd St. Stay on W 22nd St until you reach S Kinney Rd. Turn left on S Kinney Rd, which will lead you directly to the Arizona-Sonora Desert Museum. The ride is approximately 7 miles and can take around 35-45 minutes, depending on your pace.
