If you are starting from Osaka, take the Nankai Koya Line from Namba Station to Koyasan Station. The journey takes approximately 1 hour and 30 minutes and costs around 1,300 yen. Once you arrive at Koyasan Station, take the Nankai Koyasan Bus (Route 1) towards Koyasan. After about a 20-minute ride, get off at the 'Okunoin-mae' stop. From there, it's a short 10-minute walk to the Tokugawa Clan Mausoleum.

If you are driving, head to the Koyasan area via National Route 371. From Osaka, take the Hanshin Expressway to the route leading to Koyasan. The drive takes about 1 hour and 45 minutes depending on traffic. Once you reach Koyasan, follow the signs for the Tokugawa Clan Mausoleum. There is parking available near the mausoleum, but it can be limited, so plan your visit accordingly.

From Kyoto, take the JR Nara Line to Nara Station, then transfer to the Kintetsu Nara Line and take a train to Hashimoto Station. From Hashimoto, board the Nankai Koya Line directly to Koyasan Station. After arriving at Koyasan Station, take the Nankai Koyasan Bus (Route 1) towards Koyasan and get off at 'Okunoin-mae' stop. The mausoleum is a 10-minute walk from there. The total travel time is about 2.5 hours and costs around 2,500 yen.
