If you are driving from Jacksonville, take I-95 South to exit 260B for US-41 S toward Tampa. Continue on US-41 S for about 90 miles. Once you reach Osprey, keep an eye out for 401 N Tamiami Trail. Historic Spanish Point will be on your right. There is a parking lot available on-site, and parking is free.

Once in Sarasota, transfer to the Sarasota County Area Transit (SCAT) bus route 99, which will take you to Osprey. Get off at the stop closest to 401 N Tamiami Trail.
