Walking
From the center of Provincetown, head southeast on Commercial Street toward Standish Street. Turn left onto Standish Street and continue until you reach Bradford Street. Here, turn right and walk until you reach the traffic lights. Cross the intersection and continue straight onto Route 6. Follow Route 6 for approximately 10 miles. You will see signs for the Mass Audubon Wellfleet Bay Wildlife Sanctuary on your right. The entrance is located at 291 US-6, South Wellfleet, MA 02663.

If you have access to a bike, head out from the center of Provincetown on Commercial Street and ride towards the east. Continue straight until you reach Standish Street. Turn left at Standish Street, then take a right onto Bradford Street. Follow Bradford Street until you reach the traffic lights and turn right onto Route 6. Keep biking along Route 6 for about 10 miles, and you will see the Mass Audubon Wellfleet Bay Wildlife Sanctuary on your right at 291 US-6, South Wellfleet, MA 02663.

From Provincetown, you can take the Cape Cod Regional Transit Authority bus (Route 2) that heads towards Wellfleet. You can catch the bus at the nearest bus stop along Commercial Street.
