If you are starting from downtown Minneapolis, head to the Target Field Station and board the METRO Blue Line light rail towards Mall of America. Ride for 4 stops and disembark at the 'U.S. Bank Stadium' station. Transfer to the METRO Green Line light rail towards St. Paul and ride for 2 stops until you reach the 'University of Minnesota' station. From there, exit the station and walk southeast on Washington Ave SE for about 10 minutes until you reach SE Main St, where St. Anthony Main is located at 219 SE Main St.

From downtown Minneapolis, you can catch the Metro Transit bus route 3 at the Nicollet Mall. Board the bus heading towards the University of Minnesota. After about 15 minutes, get off at the stop on SE Washington Ave SE. Walk east for about 5 minutes until you reach the intersection with SE Main St. Turn left and walk north for a couple of blocks, and you will find St. Anthony Main at 219 SE Main St.
