Walking
From the Sistine Chapel, exit the Vatican Museums and head towards the entrance gate. Once outside, turn left and walk along Viale Vaticano until you reach Piazza San Pietro. At the piazza, you will see St. Peter's Basilica in front of you. Continue straight, passing through Via della Conciliazione. At the end of this street, you will reach the Tiber River. Cross the river using the Ponte Sant'Angelo. Once across, continue straight on Lungotevere Castello until you reach Via della Conciliazione. Turn left and follow the street until you reach Piazza Navona. From there, take Via del Teatro Pace and follow it until you reach Corso Vittorio Emanuele II. Turn right and then take a left onto Via del Plebiscito, which will lead you to Via dei Fori Imperiali. The Colosseum will be directly ahead as you approach.

Public Transport
From the Sistine Chapel, exit the Vatican Museums and walk to the Ottaviano metro station, which is about a 15-minute walk. Head towards the metro station on Via Ottaviano, and board Line A (the red line) towards Anagnina. Ride for two stops and get off at Termini station. From Termini, transfer to Line B (the blue line) and ride towards Laurentina. Get off at the Colosseo station. Exit the station, and you will see the Colosseum right in front of you.
