Walking
From the Sistine Chapel, exit and head towards the main entrance of the Vatican Museums. Once outside, turn left and walk along the Viale Vaticano until you reach the main road, which is Via di Porta Angelica. Continue straight until you reach the intersection with Via della Conciliazione. Cross the street and continue straight until you reach St. Peter's Square. From St. Peter's Square, head towards the right side of the square and take Via della Conciliazione until you reach the Tiber River. At the riverbank, turn right and walk along the river for a short distance until you reach the Ponte Sant'Angelo bridge. Cross the bridge and continue straight until you reach Piazza Navona. From Piazza Navona, walk east towards Via del Corso, then head north until you find the intersection with Via Barberini. Piazza Barberini will be at the end of this road.

From the Sistine Chapel, walk towards the entrance of the Vatican Museums. Exit and turn left onto Viale Vaticano. Walk until you reach the nearest bus stop, which is 'Piazza San Pietro.' Take Bus 64 in the direction of 'Termini.' Get off at 'Piazza Barberini,' which is about a 10-minute bus ride. The bus will drop you off directly at the Piazza Barberini.
